Spartacus: Vengeance is the third installment in the Spartacus series, and the official second season after its prequel, Spartacus: Blood and Sand. Spartacus: Vengeance premiered on January 27, 2012, and concluded on March 30, 2012.
It was followed up with a third, and final season, Spartacus: War of the Damned, on January 25th, 2013.

Spartacus: Vengeance takes place 6 weeks after Spartacus: Blood and Sand. It follows the story of Spartacus, (who is now portrayed by Liam McIntyre, replacing the late Andy Whitfield) after he and his fellow gladiators murder their master, Batiatus. They must fight Claudius Glaber and his troops, who have been sent to travel to Capua in order to defeat the freed gladiators and slaves. Historically, Vengeance focuses on the time from Spartacus leaving the ludus to the inevitable battle at Mount Vesuvius.

On the heels of the bloody escape from the House of Batiatus that concluded "Spartacus: Blood and Sand", the gladiator rebellion continues and begins to strike fear into the heart of the Roman Republic in "Spartacus: Vengeance". Gaius Claudius Glaber and his Roman troops are sent to Capua to crush the growing band of freed slaves that Spartacus leads before it can inflict further damage. Spartacus is presented the choice of satisfying his personal need for vengeance against the man that condemned his wife to slavery and eventual death or making the larger sacrifices necessary to keep his budding army from breaking apart. Containing all of the blood-soaked action, exotic sexuality, and villainy and heroism that has come to distinguish the series, the tale of Spartacus resumes in epic fashion.

- Liam McIntyre as Spartacus – a Thracian slave who became a gladiator, and champion, in the ludus of Quintus Batiatus. After the massacre at the House of Batiatus, Spartacus formed his own army and set on to free the slaves of Rome.
- Peter Mensah as Oenomaus – Formerly Doctore; Batiatus' taskmaster and trainer of gladiators, now a trusted mentor to Spartacus and his rebel army.
- Manu Bennett as Crixus – 'The Undefeated Gaul', he was Batiatus' top gladiator before Spartacus' arrival, now the second-in-command of Spartacus rebel army, fighting to find his lost lover, Naevia.
- Katrina Law as Mira – a slave who aided in Spartacus's escape. Now a fierce fighter for Spartacus as well as his love interest.
- Dustin Clare as Gannicus – a former champion gladiator from the House of Batiatus who was released as a free man some years before Spartacus' arrival.
- Cynthia Addai-Robinson as Naevia – a slave who was banished from the ludus before the revolt and is the love interest of Crixus.
- Dan Feuerriegel as Agron – a German gladiator who was sold to Batiatus' ludus, he was the first to join Spartacus in his revolt, where his brother Duro died. He is now one of Spartacus' trusted warriors.
- Brooke Williams as Aurelia – widow of Varro, Spartacus' only real friend in the ludus, now on the run with Spartacus' rebel army.
- Heath Jones as Donar – a German rebel and former gladiator in the House of Batiatus.
- Peter McCauley as Lucius Caelius – a Roman hermit who later joins the rebel's cause.

- Craig Parker as Gaius Claudius Glaber – Ilithyia's husband and a praetor. Responsible for Spartacus' enslavement as a gladiator and the death of his wife, he is now tasked with putting down the revolt.
- Lucy Lawless as Lucretia – Quintus Batiatus' widow, she was seriously injured from the wound she suffered at the hands of her former lover Crixus. Her recovery was presented to the people of Rome as Lucretia's personal connections with the gods, allegedly garnering her the gift of prophecy.
- Viva Bianca as Ilithyia – The daughter of Senator Albinius and wife of Glaber, now pregnant. Once a trusted friend of Lucretia before being betrayed soon before the revolt.
- Brett Tucker as Publius Varinius – Glaber's chief political rival and fellow praetor.
- Luke Pegler as Marcus – Glaber's young friend who joins him as his military tribune in order to put down the revolt.
- Paul Glover as Salvius - one of Glabers soldiers. He is fiercely loyal to his commander and desires to put down the revolt.
- Hanna Mangan-Lawrence as Seppia – a Roman noble of Capua and younger sister to Seppius.
- Tom Hobbs as Seppius – a young Capua citizen of note. He wishes to strip the honor of capturing Spartacus from Glaber in order to advance himself within the social heirarchy of Rome.
- Jason Hood as Cossutius – a Roman noble with immoral ideals.
- Greg Ward as Mercato – a Roman noble who is in charge of organizing the gladiatorial games in Capua.
- Timothy Raby as Gallienus – a Roman Magistrate of Capua. He takes over after the death of Calavius.
